Set the desired incubation para- meters on the TFT display. For a perfect temperature setting, use Control Sensor T S1 directly in the culture dish.Wait until all the incubation values have been reached. Avoid drifts simply by allowing a 2- to 3-hour warm-up period.

Place your sample under the microscope and select the cell that is to be observed. Avoid phototoxic effects by minimizing exposure to light, even during preparation.Now start your time lapse experi- ment. Avoid bright ambient light to ensure detection of weak fluo- rescence signals. For extremely exacting experiments, select Incubator XL DARK S1.

Perform manipulations that may be required for your application, e.g. temperature changes, micro- manipulation, FRAP, uncaging, ablation, etc.Analyze your experiment using one of the various modes of analysis in AxioVision. Benefit from the possibility of analysis during the experiment, for example with the "Physiology" module.
