The problem
When working with multiply stained samples (e. g. DAPI, FITC, Rhodamine) the problem is to get the true information accordingly, when trying to separate the emission of each fluorescence. |
| |
The solution
The LSM 710 provides the capability to change the settings for lasers and detectors (a single track) very rapidly by using the digital signal processor (DSP). So one line is used to scan with settings for the first dye, the next line for settings of a second dye and so on. No simultaneous detection, no cross talk at all. |
| |
Flexible definition
Up to four tracks can be defined, each with different settings for lasers and at most four channels. A channel might be any detector or the ratio of two other channels. While running a Multitrack scan the recorded channels can be displayed separately or as superimposed images. |

The simultaneous recording of
both channels at a high laser intensity causes considerable penetration of the FITC signal
into the other channel ("cross talk"). | Multitracking record of the same specimen,
with equal laser intensity: The channels are perfectly separated, without any cross talk. |
| |
| | |
Exact signal relation
Higher acquisition speed and more precise positioning of the signals in relation to each other compared to sequential imaging.
| | |
Gain speed with DDS
Signal recording during all moving directions of the scanner doubles the speed for image acquisition (Dual direction scan, DDS). The change between tracks happens at each change of the moving direction of the scanner.
| | |
Combined functionality
Correct guess: Multitracking can be combined with the scanning of Real ROIs.
