Ion Imaging
Capabilities offered by the LSM software:
  • Online and offline ratio for ratiometric dyes
  • Online and offline F/F0 for single-wavelength dyes
  • Calibration for single-wavelength and ratiometric dyes in situ and in vitro, including background correction after titration with various curve fittings according to Grynkiewicz
  • Interactive scaling of image data series
  • Interactive graphic display of the data measured in ROIs
  • Free adaptation of the lines, areas or volumes to be displayed to the sample dimensions and orientation
  • Enhanced dynamics and sensitivity compared to conventional multi-channel image acquisition, thanks to Emission Fingerprinting
  • Up to 77 frames/s (512 x 32 pixels) or 2600 lines/s with LSM 710
  • Up to 1000 frames/s (512 x 50 pixels) or more than 60,000 lines/s with LSM 7 LIVE
  • Piezoelectric objective focusing for XYZ or XZ stacks providing high time resolution in 3D
Example:
Part of the salivary gland of a fly whose secretory cells were loaded with the Fluo-4 calcium indicator.
Changes of calcium concentration in part of the tubular salivary gland displayed as F/F0 by means of pseudocolors, with blue and red representing low and high calcium, respectively.

Experiment: Stimulation of secretory activity by adding serotonin, a hormone
Result: Serotonin induces complex dynamic changes in the concentration if intracellular calcium ions. Waves of elevated calcium are passing through adjacent cells.

Solutions with different concentrations of free Ca2+ ions and the Ca2+ sensor Indo-1. Comparison of signals obtained with band pass filters and Emission Fingerprinting, with subsequent ratioing of the individual signals.
